There's 106 miles to Chicago. We've a full tank of gas, half a packet of Cigarettes, it's dark and we're wearing sunglasses.
Hit it!
Hit it!

I cannont count, how many dozen's time I've watched that great movie.
I should find and scan the pic of one of my cars, I've re-painted as the original Bluesmobile. Unbelievable, how the people have looked, when I drove that thing.

It would be a toss-up between these three:
The DeLorean from Back to the Future
The Shelby GT500 from Gone in 60 Seconds
The Suburban from Twister

@antp and taxiguy: yes, a K 70. Made in 1973, four doors, four round frontlamps and brown skai-leather - this had be to enough to be similar to a Dodge Monaco.
And: I got the car for free. At that time I had no money to spend it on cars. And a real Dodge Monaco was never easy to get in Germany, because it ws never imported.

A 1973 VW K 70? You mean like this one? A little small to be a Monaco, don't you think?

Good catch, my "Blues Mobile"-K 70 even had orignally the same colour.
Surely, it wasn't "American" enough - but it was cheap fun for me.

I diddn't just ponder this question, I am building my dream car. It's in primer right now but after a nice coat of black it'll be Burt's T/A from the first Smokey and the Bandit! Next to my T/A is my friend Brian's General Lee (season one replica).

As CCF and others have said it's a tough choice. I suppose the ones we'd want are based on various reasons for wanting them. Do you see this car?
http://www.imcdb.org/vehicle_9961-Chevrolet-Impala-1964.html
I may not be interested in a 1964 Chevy Impala Low Rider with dingle-balls around the windows, but back when I had my 1980 Plymouth Volare and my 1985 Chevrolet Caprice, I thought the grilles of them would've been perfect for the same speakers you see there. One would be for a multi-sound effects musical horn, and the other would be for a P.A. system.

Even before I started reading others' responses, I immediately decided on the the silver Aston Martin DBS in CASINO ROYALE. Others include the ML320 in THE LOST WORLD, the LeMans from NIGHT OF THE LIVING DEAD (68), the red Mercury Cougar from ON HER MAJESTY'S SECRET SERVICE. The list goes on, but the question was which "one"...

Hmmm, I think the "Bellini 3.6" (Aston DB3s) from School for Scoundrels is on the list.

If you were able to buy any car from a movie, what would you buy? This is something I would have to think about for a little bit so I'll pop back in later.
well i searched this place for the rover p6 v8 3500 driven by vinnie jones in the film lock stock and two smoking barrels as thats the car i bought about 5 years ago it sits now in my garden falling to bits a shame really

A movie car for sale:
http://cgi.ebay.de/VOLKSPOLIZEI-WARTBURG-TOURIST-DDR-OLDTIMER-VP-OSTALGIE_W0QQitemZ290246723380QQihZ019QQcategoryZ29750QQssPageNameZWDVWQQrdZ1QQ- cmdZViewItem
They wrote, that it was used for more than one production, but it's not said, for what ones (perhaps we can find out over the database).
And it's also not said clearly, if it's a real original police-car or just a later "pimped" one.
P.S. The license-plate is a fake.

I think, this is rarer than a movie-car, a theater(!)-car:
